I have a link on my desktop to a folder called ‘Programs’.
When I click the link, I would like DO to open that folder and show the sub-folders.
How can I do that?

But, is there a way to do that and hide all other drives in the tree as well? I just want the 'Programs' folder and sub-folders to be showing in the tree.

You should be able to do most of this using a Layout. Set up a lister how you want it, then save a Layout. Then go to Preferences - Listers - Layouts and drag the layout to your desktop to create a shortcut to it.
When you say you want to hide the other folders from the tree, do you mean in the Folder Tree panel, or in the file display (which is a bit like a tree itself when Flat View (Grouped) is turned on)?
If you mean the Folder Tree then the only options you have are those in Preferences - Listers - Folder Tree (e.g. set Start Folder Tree at current drive), and changing those settings will affect all Opus windows.
If you mean the file display then the starting folder is easy to change there.
