I'm demoing the Advanced FTP feature, and trying to connect to a site that uses Two Factor Authentication (SFTP-SSH besides that). In Filezilla I've used their Interactive Logon Type for this, but I'm not seeing any similar options for Directory Opus.
If it just means being prompted for a password which is different each time you use it, choosing the option to always be prompted for a password should work. (Although I'm unable to test that as I don't have a 2FA SFTP server.)
Although you may run into issues, the same as in FileZilla, with new connections being opened and closed to do things in parallel (e.g. read a folder while transferring a file). Based on that FileZilla thread, I'm not sure 2FA is a great choice if intending to use interactive (S)FTP clients. For SSH, using public key authentication may be a better choice, if it is allowed (e.g. regulatory requirements within an organisation).
I don't think that will work, unfortunately. I have a consistent user name and password, which once validated sends a token to my phone and triggers a 2nd authentication window in the ftp program asking for the token.
The decision for 2FA is firmly above my pay grade and final.
