Opus hotkeys don't work when Opus isn't running. There's no option that can change this.

You can keep Directory Opus open even when the last lister is closed by making sure this option does not have a check mark in it:

Preferences / Launching Opus / Startup / Shutdown Directory Opus when the last Lister closes
