Hi Folks,
Since I installed the latest VISTA 32 update,
the Explorer replacement function does not work anymore.
I've uninstalled and re-installed Directory Opus 9.1.1.8
two times, but it still does not work.
Does anybody know how to fix that?

Do you mean Vista SP2 or something else?
I've got all the updates that are on Windows Update on my 32-bit Vista machine and Explorer Replacement still works, but those updates don't yet include SP2.
Just SP1, but the updates I stalled that day were
- security update for Vista (KB958690)
- update for Outlook 2007 junk email filter
- security update for Vista (KB960225)
- Windows Mail junk email filter
- Update for Vista (KB959772)
- MS .Net Framework 3.5 SP1
- Malicious Software Removal Tool (March 2009).

I've got all of those installed as well without it causing any problems.
If you right-click a folder in Explorer does the Open in Directory Opus option appear? Is it bold (default)?
Either way, try turning Explorer Replacement off in Opus via Preferences, Apply or OK the change, then go back to Preferences and do the opposite. That may repair the registry values.
