Just updated to DO 12 / Version 12.22 x64 (Build 7593) but noticed this issue prior to update.
Basically the Search Boxes are not providing any results. The yellow 'location not indexed... so will take longer' message flashes for a second, vanishes and then nothing happens.
How can I fix this please.

That search box (top right of the lister) goes via Windows Search.
What happens if you do the same search from the same folder in File Explorer?
In Opus, you can also search for things without using Windows Search by selecting Tools > Find Files.

Restarted PC and now the search boxes work as expected.
