Hello,
is it possible to create a hotkey in dopus which can show and hide "hidden files" and/or "system files"?
Sometimes I need this files to show but for the better overview it would be better that they are hidden.
Any hint?

You're probably interested in the new to Opus v10 Global hide filter options under Prefs->Folders->Folder Display... i.e. enable the options there that make sense for your desired default view state. You can then use the new associated commands like:
Set HIDESYSTEMFILES=Toggle
Set GLOBALHIDEHIDDEN=Toggle
...in order to unhide hidden/system files as desired.
If you want keys to toggle the two settings individually, there are already menu items for them in the default toolbars. You can add hotkeys to them by using Settings -> Customize then editing each item (double-click it, or right-click it and select edit once in Customize mode):
