Are you behind a firewall that blocks web access? If so you will have to configure Opus to use a web proxy which can be done in Settings -> Preferences - Miscellaneous - Internet Settings.
Are you using Opus at home or in an office? If you're at home then you probably don't need to use a proxy. If you're in an office then you probably do. Either way you can find out by looking at Internet Explorer's settings, or asking a network administrator for the settings.
To check the proxy settings in IE:
On the Tools menu in Internet Explorer, click Internet Options, click the Connections tab, and then click LAN Settings.

This is for home use... I use my memory stick with DO for the office but I don't need internet access there.
I've checked that in IE (does it matter that I use Firefox...?) and all the options are unchecked.
You should see something like this printed on the screen:

If you see an error message instead then there's some kind of firewall/proxy at work somewhere. (Unless the GPSoftware web server happens to be down at the time... Try pinging another address like news.bbc.co.uk as well to be sure.)
Just to clarify, this is a home PC, not a work one.
Anyway, this is what I get from gpsoft...

I got a message from my firewall asking for access to gpsoft which I permanently granted.
I also get a similar result from the BBC.
Since this I have attempted to "Check for Updates" again but it still will not work.
Disabling it made no difference. Like I said, it's no biggie, but if there's a log file or anything else I can check, I'd be willing to dig deeper on this.
